Toxicity Evaluation

Toxicity evaluation is the systematic assessment of whether a chemical, biological agent, or environmental exposure can cause harmful effects in living organisms. It examines how factors such as dose, exposure duration, route of exposure, and biological susceptibility influence responses, often by measuring dose-response relationships and endpoints including cell viability, organ function, behavior, or reproduction. In biology, toxicity evaluation supports the identification of hazardous substances, comparison of relative risks, and investigation of mechanisms underlying cellular and organismal injury. Its findings guide drug development, environmental monitoring, chemical safety decisions, and the design of safer interventions while helping define exposure limits and priorities for further research.

Source: Chen, Y., et al. Evaluating Virulence and Pathogenesis of Aeromonas Infection in a Caenorhabditis elegans Model. J. Vis. Exp. (2018).This video demonstrates the C. elegans Liquid Toxicity Assay with Aeromonas, a method used to evaluate bacterial virulence by measuring worm survival in a nutrient-limited liquid environment. The assay reveals strain-specific pathogenicity based on the extent of toxin-induced damage and mortality in C. elegans.

2017

This work describes a new protocol to assess the ecotoxicity of pollutants, including emerging contaminants such as nanomaterials, using the marine bacterium Vibrio anguillarum. This method allows for the determination of the LC50, or mortality, the concentration that causes a 50% decrease in bacteria culturability, after a 6 h exposure.

Evaluation of the Efficacy And Toxicity of RNAs Targeting HIV-1 Production for Use in Gene or Drug Therapy

Methods to evaluate the efficacy and toxicity of RNA molecules targeting post-integration steps of the HIV-1 replication cycle are described. These methods are useful for screening new molecules and optimizing the format of existing ones.

Evaluating the Neuroprotective Effects of a Test Compound Against Alpha-Synuclein Toxicity in Rat Embryonic Neurons

2025

Start with a primary midbrain culture from rat embryos, containing dopaminergic neurons that overexpress mutated alpha-synuclein.Replace the media in the treatment wells with fresh media containing the test compound, and that in the control wells with plain media. Then incubate. In the control, alpha-synuclein forms toxic aggregates in dopaminergic neurons, causing neurite retraction and leading to neuronal death. In the treatment, the test compound interacts with alpha-synuclein, prevents its...

Rapid Evaluation of Toxicity of Chemical Compounds Using Zebrafish Embryos

Zebrafish embryos are used for evaluating the toxicity of chemical compounds. They develop externally and are sensitive to chemicals, allowing detection of subtle phenotypic changes. The experiment only requires a small amount of compound, which is directly added to the plate containing embryos, making the testing system efficient and cost-effective.
