Viral Dna Synthesis

Viral DNA synthesis is the biological process by which DNA viruses produce copies of their genomes, enabling genome maintenance, replication, and the formation of new infectious particles. It relies on template-directed DNA polymerization, in which viral or host DNA polymerases add complementary nucleotides to a growing strand, often beginning at a defined origin of replication and supported by proteins that unwind or stabilize DNA. Studying this process clarifies how viruses reproduce, interact with host cells, and evolve. It also supports antiviral drug development, viral genome analysis, and the design of viral vectors for research and biotechnology.

We describe the process of isolating high purity herpesvirus nucleocapsid DNA from infected cells. The final DNA captured from solution is of high concentration and purity, making it ideally suited for high-throughput sequencing, high fidelity PCR reactions, and transfections to produce new viral recombinants.

This method describes the use of click chemistry to measure changes in host cell transcription after infection with the Rift Valley fever virus (RVFV) strain MP-12. Results can be visualized qualitatively via fluorescence microscopy or obtained quantitatively through flow cytometry. This method is adaptable for use with other viruses.
