Z Disc M Line

Z discs and the M line are structural landmarks within a sarcomere, the contractile unit of striated muscle; Z discs mark its boundaries, while the M line occupies its center. During contraction, thin actin filaments anchored at each Z disc slide past thick myosin filaments organized around the M line, shortening the distance between Z discs without substantially changing filament length. Their arrangement maintains myofilament alignment and transmits force along muscle fibers, making these structures essential for understanding skeletal and cardiac muscle contraction, microscopy-based muscle analysis, and the effects of structural disruption in muscle disease.

Generation, Purification, and Characterization of Cell-invasive DISC1 Protein Species

The generation, purification and cell invasion of intracellular, cytoplasmic full length DISC1 protein aggresomes from cell cultures and of a labeled, multimeric recombinant DISC1 protein fragment in E. coli are described. Cell invasiveness is shown for recipient cells in cell culture and for neurons in vivo after stereotactical brain inoculation.

Synthesis of an Intein-mediated Artificial Protein Hydrogel

We present the synthesis of a split-intein-mediated protein hydrogel. The building blocks of this hydrogel are two protein copolymers each containing a subunit of a trimeric protein that serves as a crosslinker and one half of a split intein. Mixing of the two protein copolymers triggers an intein trans-splicing reaction, yielding a polypeptide unit that self-assembles into a hydrogel.
