Docetaxel Resistance

Docetaxel resistance is the reduced sensitivity of cancer cells to docetaxel, a taxane chemotherapy that disrupts cell division, and it can limit treatment effectiveness and disease control. Resistance develops through mechanisms such as increased drug efflux by ATP-binding cassette transporters, changes in tubulin or microtubule dynamics, altered drug metabolism, and activation of cell-survival pathways that prevent apoptosis. Studying these processes helps researchers identify molecular biomarkers, understand treatment failure, and design strategies that restore drug sensitivity. Findings support improved combination therapies, patient stratification, and the development of new approaches for managing recurrent or treatment-resistant cancers.

In this paper, we report a protocol describing an in vivo method to measure microtubule dynamic instability in docetaxel-resistant breast cancer cells (MCF-7TXT). In this method, a deconvolution microscopy imaging system is used to detect the expression of GFP-tubulin in target cells.
