Glycosaminoglycan Scaffold

A glycosaminoglycan scaffold is a biomaterial or extracellular-matrix-inspired structure built from glycosaminoglycans, linear polysaccharides that help organize tissues and regulate cell signaling. Its negatively charged chains bind water, proteins, and signaling molecules, creating a hydrated matrix that can influence cell adhesion, migration, proliferation, and communication. In cancer research, these scaffolds model features of the tumor microenvironment and provide controlled platforms for studying cancer cell behavior, extracellular-matrix interactions, and therapeutic responses. They can also support the development and evaluation of engineered tissues and drug-delivery strategies, helping connect molecular mechanisms with tumor progression and treatment design.

In this video, we demonstrate a procedure for combined Alcian blue and silver staining of sulfated glycosaminoglycan oligosaccharide fragments. This combined staining technique significantly amplifies the bands of glycosaminoglycan polymers of different lengths in polyacrylamide gels.

This report describes techniques to isolate and purify sulfated glycosaminoglycans (GAGs) from biological samples and a polyacrylamide gel electrophoresis approach to approximate their size. GAGs contribute to tissue structure and influence signaling processes via electrostatic interaction with proteins. GAG polymer length contributes to their binding affinity for cognate ligands.
