Nog Mouse Model

The NOG mouse model is a profoundly immunodeficient mouse strain used to study human cells and tumors in vivo, making it valuable for cancer research. NOD/Shi-scid IL2Rγnull mice lack functional T and B cells, have severely impaired natural killer cell activity, and cannot signal effectively through the common gamma-chain cytokine receptor, allowing human tumor cells or patient-derived tumor fragments to engraft with limited immune rejection. Researchers use NOG mice to establish xenograft and patient-derived xenograft models, evaluate tumor growth and metastasis, and assess anticancer drugs, immunotherapies, and combination treatments in a living organism.

Establishment of the Dual Humanized TK-NOG Mouse Model for HIV-associated Liver Pathogenesis

This protocol provides a reliable method to establish humanized mice with both human immune system and liver cells. Dual reconstituted immunodeficient mice achieved via intrasplenic injection of human hepatocytes and CD34+ hematopoietic stem cells are susceptible to human immunodeficiency virus-1 infection and recapitulate liver damage as observed in HIV-infected patients.

Establishing Experimental Metastases Mouse Model: Implanting GFP Expressing CRC Organoid Cells in PDX Model to Detect Micrometastases

2023

This video describes the protocol for generating metastases by the PDX derived CRC organoids labeled with GFP lentivirus into the spleen of an immunodeficient mouse. Cancer micrometastases colonizing other organs can be detected using a fluorescence microscope to assess the GFP expression.

Spontaneous Metastasis Mouse Models: A Platform to Study the Metastatic Potential of Orthotopically Injected Colorectal Cancer Cells

2023

This video describes the protocol to establish a spontaneous metastasis mouse model of colorectal cancer patient-derived xenografts or PDXs. These xenograft models partially mimic the features of human carcinomas and understand the critical aspects of human malignancies, including invasion and metastasis.

Mouse Models of Periventricular Leukomalacia

2010

We established mouse models of periventricular leukomalacia (PVL), the predominant brain injury in premature infants characterized by periventricular white matter lesions. Hypoxia/ischemia with/without systemic infection are the primary causes of PVL. Unilateral carotid ligation and hypoxia exposure with/without lipopolysaccharide injection creates PVL-like lesions in P6 mice.
