3d Scaffold Fabrication

3D scaffold fabrication is the design and production of porous, three-dimensional structures that support cell attachment, growth, and tissue formation, making it important in biomaterials research and regenerative medicine. In chemistry-based approaches, polymers or other biomaterials are shaped layer by layer or molded into defined architectures, then stabilized through physical interactions, chemical crosslinking, or controlled solidification; material composition and processing conditions determine pore size, mechanical strength, and degradation. These scaffolds provide platforms for studying cell behavior, delivering therapeutic molecules, and developing engineered tissues, while chemical control of their surfaces and networks helps tailor biological compatibility and functional performance.

Bioactive and mechanically reliable metal scaffolds have been fabricated through a method which consists of two processes, dynamic freeze casting for the fabrication of porous Ti, and coating and densification of the Ti scaffolds. The densification process is simple, effective and applicable to the fabrication of functionally graded scaffolds.

In the past many in vitro culture systems -- mainly monolayer cultures -- often suffered from the disadvantage that differentiated primary cells had a relatively short life-span and de-differentiated during culture. As a consequence, most of their organ-specific functions were lost rapidly. Thus, in order to reproduce better conditions for these cells in vitro, modifications and adaptations have been made to conventional monolayer cultures.

Scaffolds capable of fitting within cranio-maxillofacial (CMF) bone defects while exhibiting osteoconductivity and bioactivity are of interest. This protocol describes the preparation of a shape memory scaffold based on polycaprolactone diacrylate (PCL-DA) using a solvent-casting particulate-leaching (SCPL) method employing a fused salt template and application of a bioactive polydopamine coating.
