Glossopharyngeal Nerve

The glossopharyngeal nerve, or cranial nerve IX, is a mixed sensory, motor, and parasympathetic nerve that links the brainstem with the tongue, pharynx, middle ear, and carotid region. Its sensory fibers carry taste and general sensation from the posterior third of the tongue and pharynx, initiate the afferent limb of the gag reflex, and monitor blood pressure and blood chemistry through carotid sinus and body receptors; motor fibers activate the stylopharyngeus, while parasympathetic fibers stimulate parotid secretion. Clinically, examination of swallowing, gag responses, and posterior tongue sensation helps localize lesions, while glossopharyngeal neuralgia illustrates the effects of dysfunction.

Calcium is a ubiquitous messenger in the nervous system, essential for triggering neurotransmitter release and changes in synaptic strength. Here we demonstrate a technique for loading Ca2+-indicators into Drosophila nerve terminals. We also demonstrate fabrication of the required apparatus and emphasize points critical for the technique's success.
