Craniofacial Cartilage

Craniofacial cartilage is the cartilaginous tissue that shapes and supports developing structures of the face and skull, making it central to vertebrate form and function. During development, neural crest-derived mesenchymal cells condense and differentiate into chondrocytes, which produce an extracellular matrix rich in collagen and proteoglycans; coordinated cell proliferation and matrix deposition drive cartilage growth, while some regions later contribute to bone through endochondral ossification. Studying these processes helps developmental biologists understand craniofacial patterning and congenital abnormalities, while informing research on cartilage repair, tissue engineering, and regenerative medicine.

Electroporation of Craniofacial Mesenchyme

Craniofacial cartilages develop in close contact with other tissues and are difficult to manipulate in live animals. We are using electroporation to deliver molecular tools during growth of the craniofacial skeleton while bypassing early embryonic effects. This approach will allow us to efficiently test candidate molecules in vivo.

Dissection of Zebrafish Craniofacial Tissues Upon Staining with Alcian Blue

The protocol described here presents a simple and easy-to-follow procedure for staining and dissecting craniofacial cartilages in a 5-day-old zebrafish larva. It can be used to study the anatomy, shape, and size of these structures under various developmental conditions.

Cell organization of craniofacial bones has long been hypothesized but never directly visualized. Multi-spectral cell labeling and in vivo live imaging allows visualization of dynamic cell behavior in zebrafish lower jaw. Here, we detail the protocol to manipulate Zebrabow transgenic fish and directly observe cell intercalation and morphological changes of chondrocytes in the Meckel’s cartilage.
