Human Neuronal Culture

Human neuronal culture is the laboratory maintenance and differentiation of human neurons or neural progenitor cells under controlled conditions, providing a model for studying how the human nervous system develops. Cells are grown on suitable substrates in defined media, where nutrient availability, cell density, and signaling molecules influence survival, neurite extension, maturation, and synaptic connectivity. In developmental biology, these cultures allow researchers to examine neuronal differentiation, axon and dendrite formation, and responses to genetic or environmental perturbations in a controlled setting. They also support disease modeling and the investigation of developmental mechanisms that are difficult to observe directly in human tissue.

Human neuroprogenitor cells (NPCs) were expanded under proliferating conditions. NPCs were differentiated into neuron-rich cultures in the presence of a combination of neurotrophins. Neuronal markers were detected by immunofluorescence staining. To isolate a pure population of neurons, NPCs were differentiated on PEN membrane slides and laser capture microdissection was performed.
