Myocardial Renewal

Myocardial renewal is the replacement or restoration of heart muscle cells, a process that influences how the heart forms, grows, and responds to injury. During development, signals that regulate cardiomyocyte proliferation, differentiation, and maturation help build functional cardiac tissue, while limited regenerative activity may persist after birth. Researchers study these cellular behaviors, along with cardiac progenitor populations and tissue signaling, to understand why renewal declines and how it might be enhanced. This knowledge supports research into congenital heart development, myocardial repair, and regenerative therapies aimed at restoring contractile function after cardiac damage.

SC1 functions through dual inhibition of Ras- GAP and ERK1. We tested the function of SC1 in supporting mouse ES cell self-renewal in the absence of LIF and showed that SC1 is able to maintain self-renewal of mouse ES cell cultures.

The generation of aligned myocardial tissue is a key requirement for adapting the recent advances in stem cell biology to clinically useful purposes. Herein we describe a microcontact printing approach for the precise control of cell shape and function. Using highly purified populations of embryonic stem cell derived cardiac progenitors, we then generate anisotropic functional myocardial tissue.

The post-mortem assessment of myocardial infarction (MI) in rodents is based on quantification of the infarct on stained heart sections. We describe an accurate method to quantify the infarct size using systematic sampling of harvested rat hearts from base to apex and image analyses of trichrome-stained histological sections.
