High-speed Microscopy System

A high-speed microscopy system is an imaging platform that captures rapid biological, physical, or engineered events at temporal resolutions beyond conventional microscopy, making transient behavior measurable rather than inferred. It combines fast image sensors, controlled illumination, high-speed optics, and synchronized acquisition to record successive frames while limiting motion blur and preserving spatial detail. In engineering, these systems help analyze fluid motion, mechanical deformation, microfabrication processes, and dynamic material responses. Quantitative image sequences can reveal failure mechanisms, transport phenomena, and process variability, supporting the design, testing, and optimization of devices and manufacturing methods.

Recently we mapped the three-dimensional (3D) spatial locations of transport routes for various proteins translocating inside primary cilia in live cells. Here this paper details the experimental setup, the process of biological samples and the data analyses for the 3D super-resolution fluorescence imaging approach newly applied in live primary cilia.
