Outflow Medium Collection

Outflow Medium Collection is a sampling method in which liquid leaving a biological culture or experimental system is recovered for analysis, preserving information released into the surrounding environment. As medium flows through an outlet, researchers collect it under controlled, contamination-limited conditions and may process it for nucleic acids, proteins, metabolites, or other cell-derived materials. In genetics, this approach can complement direct tissue sampling by enabling analysis of extracellular DNA or RNA and other molecular signals without disrupting the source system. Collected samples support genetic profiling, biomarker studies, and monitoring of cellular responses over time.

2017

This protocol provides a method for the collection of mouse embryonic stem cell (mESC)-conditioned medium (mESC-CM) derived from serum (fetal bovine serum, FBS)- and feeder (mouse embryonic fibroblasts, MEFs)-free conditions for a cell-free approach. It may be applicable for the treatment of aging and aging-associated diseases.

The present study reports a protocol for chromosome screening of human embryos that uses spent culture medium, which avoids embryo biopsy and enables chromosome ploidy identification using NGS. The present article presents the detailed procedure, including the preparation of culture medium, whole genome amplification (WGA), next-generation sequencing (NGS) library preparation, and data analysis.

This video describes a method for purifying Wolbachia pipientis from an Anopheles gambiae cell line and then culturing the endosymbiont in cell-free medium. An assay for viability of the bacterium is demonstrated.

A Method for Generating Oligodendrocyte-Conditioned Medium

2025

This video demonstrates the production of oligodendrocyte-conditioned medium (OCM) from oligodendrocyte lineage cells by stressing them to enhance their molecular release into the medium. This medium, which is rich in growth factors, cytokines, and other bioactive molecules, can be added to neuron cultures to gain insight into the impact of oligodendrocytes-secreted factors on neuronal physiology and connectivity.
