Radiolabeled Transcript Analysis

Radiolabeled transcript analysis is a molecular biology method that uses radioactive labels to detect and characterize RNA transcripts, providing information about gene expression and RNA processing. In a typical assay, cells or transcription reactions incorporate radiolabeled nucleotides into newly synthesized RNA, which is then separated by size or sequence and visualized through autoradiography or related detection methods. In genetics, the approach can reveal transcriptional activity, transcript abundance, processing patterns, and RNA stability. Although many modern assays use fluorescent or sequencing-based detection, radiolabeled transcript analysis remains valuable for measuring RNA synthesis and examining gene regulation with high sensitivity.

Analysis of Termination of Transcription Using BrUTP-strand-specific Transcription Run-on (TRO) Approach

We describe a basic experimental approach for analysis of termination of transcription by RNA polymerase II in vivo using BrUTP by the strand-specific transcription run-on (TRO) approach in budding yeast. This protocol can be extended to study transcription termination by other RNA polymerases both in yeast and higher eukaryotes.

This manuscript describes a protocol that applies comprehensive profiling for analysis of transcriptional programs induced in specific brain nuclei of rodents following behavioral paradigms. Herein, this approach is illustrated in the context of profiling genes induced in the nucleus accumbens (NAc) of mice following acute cocaine exposure, utilizing microfluidic qPCR arrays.
