Acid-induced Cell Model

An acid-induced cell model is an in vitro system that exposes cultured cells to controlled acidic conditions to reproduce acid stress encountered in physiological or infection-related environments. Lower extracellular pH can disrupt membrane function, protein homeostasis, and cellular metabolism, triggering stress responses, inflammatory signaling, or cell injury depending on exposure intensity and duration. In immunology and infection research, this model helps examine how acidic microenvironments influence host-cell viability, barrier integrity, and immune responses, as well as how pathogens adapt to acid stress. By standardizing pH and exposure conditions, researchers can isolate acid-dependent effects and evaluate potential protective or therapeutic interventions.

We illustrate a straight-forward method to derive murine primary macrophages from bone marrow cells and a simple method to prepare BSA-fatty acid conjugates. Then we demonstrate that saturated fatty acids can induce macrophage cell death, and such cell death is positively associated with cellular accumulation of ceramide levels.

The present protocol describes a lung injury model in mice using oleic acid to mimic acute respiratory distress syndrome (ARDS). This model increases the inflammatory mediators on edema and decreases lung compliance. Oleic acid is used in the salt form (oleate) since this physiological form avoids the risk of embolism.
