Allogeneic Stem Cells

Allogeneic stem cells are stem cells obtained from a genetically different donor and transferred to a recipient, where they can rebuild damaged tissues or reconstitute the immune and blood-forming systems. In hematopoietic stem-cell transplantation, donor cells home to the recipient’s bone marrow, engraft, and generate new immune cells; compatibility between donor and recipient human leukocyte antigens helps limit rejection and graft-versus-host disease. These transplants treat leukemia, lymphoma, marrow failure, and certain immune deficiencies, while donor immune cells may eliminate residual cancer through graft-versus-leukemia effects. In immunology and infection research, they also reveal how immune reconstitution, immunosuppression, and donor-recipient interactions shape susceptibility to pathogens and transplant outcomes.

A rapid, simple and cost-effective protocol for the generation of donor-derived multivirus-specific CTLs (rCTL) for infusion to allogeneic hematopoietic stem cell transplant (HSCT) recipients at risk of developing CMV, Adv or EBV infections. This manufacturing process is GMP-compliant and should ensure the broader implementation of T-cell immunotherapy beyond specialized centers.

An experimental technique for the treatment of osteochondral defects in the rabbit's knee joint is described. The implantation of allogeneic mesenchymal stem cells into osteochondral defects provides a promising development in the field of tissue engineering. The preparation of fibrin-cell-clots in vitro offers a standardized method for implantation.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
