Anti-cd3/28 Activation

Anti-CD3/28 activation is an in vitro method for stimulating T cells through their antigen receptor and essential costimulatory pathway, enabling controlled analysis of T-cell function in immunology and infection research. Immobilized or bead-bound antibodies engage the CD3 component of the T-cell receptor complex while simultaneously ligating CD28, triggering signaling pathways that promote T-cell receptor activation, proliferation, cytokine production, and differentiation. This approach provides a reproducible alternative to antigen-specific stimulation and supports the expansion and characterization of primary T cells. Researchers use it to examine immune responses, model activation states, evaluate infection-related dysfunction, and generate activated cells for studies of immunotherapies and cellular therapies.

An In Vitro Assay to Evaluate the Anti-Tumor Activity of Tumor-Specific CAR T Cells

This video demonstrates an in vitro assay to validate chimeric antigen receptor (CAR) T cell activity. B-cell lymphoma cells expressing CD19 antigen and cytoplasmic luciferase are co-cultured with CD19-specific CAR T cells. CAR binding to CD19 on a tumor induces the CAR T cells to release cytokines and toxins that cause tumor cell cytotoxicity. The CAR T cell activity is evaluated by quantifying the released cytokine and decreased luciferase-mediated bioluminescence in the tumor cells.

An In Vitro Method to Determine the Anti-Apoptotic Activity of Antibodies against TNFα

This video demonstrates a technique for assessing the efficacy of antibodies against tumor necrosis factor-alpha (TNFα), an inflammatory cytokine, through an in vitro bioanalytical approach. TNFα, when introduced to stressed cells, triggers apoptosis. However, binding to the antibodies confines TNFα, averting apoptosis. The degree of apoptosis is quantified using a bioluminescence assay.
