Cd11b Cd45

CD11b and CD45 are cell-surface markers used together to identify and characterize immune cells, particularly in immunology and infection research. CD45, a pan-leukocyte protein tyrosine phosphatase, distinguishes hematopoietic cells from most nonimmune cells, while CD11b, an integrin involved in adhesion and migration, helps identify myeloid populations and related leukocyte subsets. Researchers detect these markers by antibody staining and flow cytometry or tissue imaging, using fluorescence intensity and co-expression patterns to define cell populations. This combination supports measurement of leukocyte recruitment, myeloid-cell accumulation, and immune responses during infection, inflammation, tissue injury, and experimental treatment.

Here, we present a protocol to isolate microglia from postnatal mouse pups (day 1) for in vitro experimentation. This improvised method of isolation generates both high yield and purity, a significant advantage over alternate methods that allows broad range experimentation for the purposes of elucidating microglial biology.

Source: Hidmark, A. S., et al. Analysis of Immune Cells in Single Sciatic Nerves and Dorsal Root Ganglion from a Single Mouse Using Flow Cytometry. J. Vis. Exp. (2017). This video demonstrates the use of flow cytometry to analyze immune cells in mouse sciatic nerves and dorsal root ganglia (DRGs). The process involves labeling immune cells with fluorophore- and biotin-tagged antibodies, stabilizing and permeabilizing the cells, and using fluorescent markers to detect cell-surface and nuclear...

Inflammation plays a central role in the pathogenesis of ischemic stroke. Increasing evidence suggests that it acts as a double-edged sword which exacerbates early brain injury, but also contributes to later repair. This protocol describes the isolation of immune cells from the ischemic brain and their subsequent flow cytometric phenotyping.

A protocol is reported here for the selective ablation of renal macrophages to study their regeneration using the human CD59/intermedilysin cell ablation tool. This method is also applicable for studying the function and regeneration of the other cell populations in the kidney, liver, and fatty tissue.
