Coupon Harvesting

Coupon harvesting is the controlled retrieval of standardized material samples used to collect and study microbial biofilms, making it useful for investigating surface-associated infection. Coupons are placed in a defined biological or environmental system, where microorganisms attach, multiply, and produce an extracellular matrix; researchers then remove the coupons at selected time points for microscopy, culture, or molecular analysis. In immunology and infection research, this approach helps characterize pathogen colonization, community development, and responses to antimicrobial treatments under reproducible conditions. Comparing harvested coupons can reveal how surface properties, environmental conditions, or interventions influence biofilm formation and persistence.

Burch in their book The Principles of Humane Experimental Technique. The 3 R's are replacement, reduction, and refinement of the use of animals in research. The use of cell lines and tissue cultures that originated from research animals is a replacement technique, as it allows for many experiments to be conducted in vitro. Harvesting...

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN Many animal experiments rely on final data collection time points that are gathered from the harvesting and testing of organs and tissues. The use of appropriate methods for the collection of organs and tissues can impact the quality of the samples and the analysis of the data that is gleaned for the testing of the tissues. The method of euthanasia of the animal can also impact the quality of...

Marginating-hepatic leukocytes exhibit unique characteristics and distinct immunological functions compared to other leukocyte populations. Here we describe a method for selective harvesting of this specific hepatic cell population, through forced perfusion of the liver of rats or mice. Marginating-hepatic leukocytes seem critical in determining susceptibility to hepatic-related diseases and metastases.
