Macrophage Accumulation

Macrophage accumulation is the recruitment and concentration of macrophages within tissues, where these innate immune cells detect threats, remove damaged material, and coordinate inflammation. During infection, microbial products and signals from injured or activated cells stimulate chemokine and cytokine release, guiding circulating monocytes into affected sites, where they differentiate into macrophages and respond through phagocytosis, antimicrobial activity, and cytokine production. This process can help contain pathogens and support tissue repair, but persistent accumulation may sustain inflammation and contribute to tissue damage. Studying macrophage accumulation helps researchers understand host defense, infectious disease progression, inflammatory pathology, and potential immune-targeted therapies.

Investigation of Macrophage Polarization Using Bone Marrow Derived Macrophages

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.

A protocol is presented for cell culture of macrophage colony-stimulating factor (M-CSF) differentiated human monocyte-derived macrophages. The protocol utilizes cryopreservation of monocytes coupled with their bulk differentiation into macrophages. Then harvested macrophages can then be seeded into culture wells at required cell densities for carrying out experiments.

The protocol presented here describes methods for depleting macrophages in vivo, deriving polarized macrophages from bone marrow aspirates, and adoptively transferring macrophages into mice.
