Proliferation Assessment

Proliferation assessment is the measurement of how rapidly and extensively cells increase in number, providing a quantitative view of growth, activation, and response to external stimuli. In immunology and infection research, it commonly tracks cell division through DNA synthesis, incorporation of nucleotide analogs, expression of proliferation-associated markers, or dilution of fluorescent dyes as cells divide. These measurements help characterize lymphocyte activation, evaluate immune responses to antigens or pathogens, and determine how infectious agents or treatments alter host-cell growth. Proliferation data can therefore clarify cellular mechanisms of immunity, support vaccine and therapeutic research, and reveal changes associated with infection or disease.

Here, we describe a protocol for generating apical-out intestinal organoids from standard matrix-embedded organoid cultures. It also outlines the subsequent incorporation of EdU into actively proliferating cells and the semiautomatic quantification of EdU-positive cells.

In this video, we determine the proliferative capacity of neuronal progenitor cells by measuring the incorporation of radioactive tracer tritiated thymidine into the DNA of cells. This technique can be used to study cell proliferation after treatment with certain drugs and growth factors.
