Thymocyte Isolation

Thymocyte isolation is the process of separating developing T cells, or thymocytes, from thymic tissue for study of immune development and function. It typically involves mechanically dissociating the thymus into a single-cell suspension, removing tissue debris by filtration, and enriching viable cells through washing or density-based separation under controlled conditions. Isolated thymocytes provide a useful model for examining T-cell maturation, selection, proliferation, apoptosis, and responses to infection or immune-modulating signals. The resulting cell preparations support flow cytometry, molecular analyses, and ex vivo assays that clarify how the thymus generates a diverse and functional T-cell repertoire.

In this video, we describe a method to identify the small-molecule kinase inhibitors that modulate the apoptosis of self-reactive CD4+CD8+ double-positive immature thymocytes. Apoptosis is induced in the double-positive thymocytes by activating them with anti-CD3- and anti-CD28-coated magnetic beads; this is followed by a small-molecule inhibitor treatment and flow cytometry analysis to detect if the inhibitors modulate the apoptotic marker expression.

Here, we present a protocol to prepare apoptotic thymocytes and peritoneal macrophages and analyze the efficiency of efferocytosis and the specific inhibitor-mediated blocking of apoptotic thymocytes engulfment. This protocol has a broad application in cell-mediated clearance of other particles including artificial beads and bacteria.

The goal of the protocol is to show longitudinal intravital real-time tracking of thymocytes by laser scanning microscopy in thymic implants in the anterior chamber of the mouse eye. The transparency of the cornea and vascularization of the graft allows for continuously recording progenitor cell recruitment and mature T-cell egress.
