C-fos Immunofluorescence

C-fos immunofluorescence is a microscopy-based method that detects c-Fos protein in fixed cells or tissue, providing a marker of recent cellular activation. The technique uses primary antibodies that bind c-Fos, followed by fluorescently labeled secondary antibodies; the resulting signal is visualized with a fluorescence microscope and localized to cell nuclei. In medicine and biomedical research, c-fos immunofluorescence helps map activated neurons, assess responses to injury or treatment, and investigate signaling pathways associated with pain, stress, inflammation, and disease. Quantifying fluorescent c-Fos-positive cells can reveal spatial and cellular patterns of activation and support studies of therapeutic mechanisms.

Here we present a Fluorescence Activated Cell Sorting (FACS) protocol to study molecular alterations in Fos-expressing neuronal ensembles from both fresh and frozen brain tissue. The use of frozen tissue allows FACS isolation of many brain areas over multiple sessions to maximize the use of valuable animal subjects.

Here, we describe protocols for the genetic and chemical validation of c-Fos and Dusp1 as a drug target in leukemia using in vitro and in vivo genetic and humanized mouse models. This method can be applied to any target for genetic validation and therapeutic development.
