Cerebrovascular Endothelium

Cerebrovascular endothelium is the specialized layer of endothelial cells lining blood vessels in the brain, where it helps maintain the blood-brain barrier and regulate communication between blood and neural tissue. Its tightly connected cells use junctional proteins, selective transporters, and signaling pathways to control molecular permeability, vascular tone, nutrient delivery, and immune-cell trafficking within the neurovascular unit. Studying this tissue is essential for understanding stroke, brain tumors, neuroinflammation, and neurodegenerative disease. It also supports the development of drug-delivery strategies that safely transport therapeutics across the blood-brain barrier while preserving cerebral homeostasis.

After puncturing the aorta through the inferior vena cava (IVC) to create an aorto-caval fistula in the mouse, solution containing a drug is infused into the IVC via the same needle, followed by incubation. This method enables more robust drug delivery to the venous endothelium compared to the external route.
