In Vivo Approaches

In vivo approaches are experimental methods that investigate biological processes, disease mechanisms, or therapeutic effects within living organisms, preserving the interactions of intact tissues and organ systems. These studies typically administer a drug, treatment, or biological intervention to an animal or human participant and then measure outcomes such as physiological responses, pharmacokinetics, toxicity, or disease progression under defined conditions. In medicine, in vivo research connects molecular and cellular findings with whole-organism function, helping evaluate treatment efficacy and safety before clinical use, refine therapeutic strategies, and assess how factors such as metabolism, immune responses, and organ interactions influence outcomes.

Here we describe a method used to isolate native protein complexes from Drosophila embryos after in vivo crosslinking followed by purification of the crosslinked complexes for subsequent structure-function analysis.

We outline a protocol that implements both in vivo and ex vivo approaches to study ovarian cancer colonization of peritoneal adipose tissues, particularly the omentum. Furthermore, we present a protocol to quantitate and analyze immune cell-structures in the omentum known as milky spots, which promote metastases of peritoneal adipose.

A Cre-Lox P Recombination Approach for the Detection of Cell Fusion In Vivo

A method to track cell fusion in living organisms over time is described. The approach utilizes Cre-LoxP recombination to induce luciferase expression upon cell fusion. The luminescent signal generated can be detected in living organisms using biophotonic imaging systems with a sensitivity of detection of ˜1,000 cells in peripheral tissues.

We present a detailed method to study human placental physiology in vivo at term. The method combines blood sampling from the incoming and outgoing vessels on the maternal and fetal sides of the placenta with ultrasound measurements of volume blood flow and placental tissue sampling.

A simple, efficient and robust way to synchronize the delivery of multiple viral components to plant cells via Agrobacterium-mediated transient expression is described. This approach is amenable for studying replication, encapsidation followed by in vitro reassembly of non-viral components into genome depleted optical viral ghosts suitable for biomedical applications.
