Interstitial Fibrosis

Interstitial fibrosis is the abnormal accumulation of extracellular matrix, particularly collagen, within the connective tissue spaces between cells, where it can disrupt organ structure and function. Persistent injury or inflammation activates fibroblasts and myofibroblasts, which produce excess matrix and alter tissue architecture; profibrotic signals such as transforming growth factor beta help sustain this response. In medicine, interstitial fibrosis is a key feature of chronic diseases affecting the lungs, kidneys, liver, and heart. Assessing its distribution and severity supports diagnosis, prognosis, treatment selection, and evaluation of antifibrotic therapies aimed at limiting irreversible scarring and preserving organ function.

Interstitial fluid flow is elevated in solid tumors and can modulate tumor cell invasion. Here we describe a technique to apply interstitial fluid flow to cells embedded in a matrix and then measure its effects on cell invasion. This technique can be easily adapted to study other systems.
