Muscle Dissection Protocol

A muscle dissection protocol is a structured method for exposing, separating, and examining muscle tissue and its associated anatomical structures, providing a foundation for understanding human form and function. The procedure typically follows anatomical planes while careful incision and blunt separation reveal muscle origins, insertions, fiber orientation, fascia, nerves, and blood vessels without unnecessarily damaging neighboring tissues. In medicine, muscle dissection supports anatomy education, surgical planning, radiologic interpretation, and investigation of neuromuscular disorders or traumatic injury. Accurate documentation of observed structures also strengthens anatomical knowledge and helps connect gross morphology with movement, innervation, and clinical signs.

Drosophila flight muscle is a powerful model to study transcriptional regulation, alternative splicing, metabolism, and mechanobiology. We present a protocol for dissection of fluorescent-labeled flight muscle from live pupae to generate highly enriched samples ideal for proteomics and deep-sequencing. These samples can offer important mechanistic insights into diverse aspects of muscle development.
