Point Of Sickling

Point of sickling is the oxygen tension at which red blood cells containing sickle hemoglobin begin to adopt a sickle shape, making it an important measure of hemoglobin S behavior in sickle cell disease. As oxygen levels fall, deoxygenated hemoglobin S molecules associate into rigid polymers that distort red blood cells, reducing their flexibility and potentially obstructing small blood vessels. Laboratory assessment of the point of sickling helps characterize the tendency of blood to sickle under low-oxygen conditions and supports investigation of disease severity, hemoglobin variants, and antisickling treatments. The measurement also clarifies how oxygen availability influences clinical complications such as vaso-occlusion and tissue hypoxia.

We have outlined a method of continuous manual exchange transfusion for the treatment of sickle cell disease in patients. This safe protocol was designed to effectively limit iron overload in patients in need of chronic transfusions and can be used extensively without any special equipment.

Characterization of Sickling During Controlled Automated Deoxygenation with Oxygen Gradient Ektacytometry

Here, we present oxygen gradient ektacytometry, a rapid and reproducible method to measure red blood cell deformability in samples from patients with sickle cell disease under controlled deoxygenation and reoxygenation. This technique provides a way to study red blood cell sickling and to monitor sickle cell disease treatment efficacy.

Synthesis of Masarimycin, a Small Molecule Inhibitor of Gram-Positive Bacterial Growth

A detailed protocol is presented for preparing the bacteriostatic diamide masarimycin, a small molecule probe that inhibits the growth of Bacillus subtilis and Streptococcus pneumoniae by targeting cell wall degradation. Its application as a chemical probe is demonstrated in synergy/antagonism assays and morphological studies with B. subtilis and S. pneumoniae.

Magnetic Levitation Coupled with Portable Imaging and Analysis for Disease Diagnostics

We present a magnetic levitation technique coupled with automated imaging and analysis in both a smartphone-compatible device and a device with embedded imaging and processing. This is applied to measure the density distribution of cells with two demonstrated biomedical applications: sickle cell disease diagnosis and separating white and red blood cells.
