Astrocyte Immunostaining

Astrocyte immunostaining is a laboratory method that uses antibodies to identify and visualize astrocytes, star-shaped glial cells that support neuronal function and maintain the brain’s extracellular environment. In this process, tissue is fixed, permeabilized, and exposed to primary antibodies that bind astrocyte-associated proteins, followed by labeled secondary antibodies that generate a fluorescent or chromogenic signal. Researchers use the resulting staining patterns to examine astrocyte distribution, morphology, and changes in neural tissue. The technique supports studies of brain development, neuroinflammation, injury, neurodegenerative disease, and interactions between glial cells and neurons in neuroscience.

(2015).This video demonstrates the imaging of fluorescently labeled astrocytes in a mouse brain section using a confocal microscope. The brain section is positioned under the appropriate objective lens, and suitable fluorophores and filters are selected. After optimizing the exposure automatically, the scanning parameters are defined to ensure high-resolution 2D images. Z-stack...

Astrocytes are one of the most important key players in the central nervous system (CNS). Here, we are reporting a practical method of sexed hippocampal astrocyte culture protocol in order to study the mechanisms underlying the astrocyte function in male and female neonate pups after in-vitro ischemia.
