Cd11b Microglia

CD11b microglia are central nervous system-resident immune cells identified by expression of CD11b, an integrin associated with myeloid-cell adhesion and immune function. CD11b forms complement receptor 3 with CD18, enabling microglia to bind iC3b-opsonized targets and support migration, engulfment, and phagocytic clearance. In neuroscience research, CD11b immunolabeling, flow cytometry, and magnetic isolation help detect and quantify microglia in brain tissue and assess responses to injury, infection, or neurodegeneration. These approaches support studies of neuroinflammation, although CD11b should be interpreted with additional markers because peripheral myeloid cells can share this phenotype.

Here, we present a protocol to isolate microglia from postnatal mouse pups (day 1) for in vitro experimentation. This improvised method of isolation generates both high yield and purity, a significant advantage over alternate methods that allows broad range experimentation for the purposes of elucidating microglial biology.

In this video, we describe a protocol for isolating CD11b cells, including microglia, from mouse pup brains using magnetic-activated cell sorting. The brain tissues are homogenized to obtain single-cell suspensions, which are incubated with anti-CD11b antibody-coated microbeads, followed by a magnetic column-based separation to purify the CD11b+ cell population.

We outline methods for the efficient and quick isolation/culture of viable microglia from the neonatal cerebral cortex and adult spinal cord. The dissection and plating of cortical microglia can be accomplished within 90 minutes, with the subsequent microglial harvest taking place ~ 10 days following the initial dissection.

Microglia (immune cells of the brain), are used as a surrogate biosensor to determine how nanoparticles influence neurotoxicity. We describe a series of experiments designed to assay microglial response to nanoparticles and exposure of hypothalamic neurons to supernatant from activated microglia to determine neurotoxicity.
