Intracellular Injection

Intracellular injection is a microscale technique that delivers a defined substance directly into a living cell, enabling researchers to manipulate or label cellular components with high spatial precision. In neuroscience, a fine glass micropipette penetrates the cell membrane and releases material through controlled pressure or electrical pulses while the neuron is maintained under microscopic observation. Investigators use this approach to introduce fluorescent dyes, tracers, nucleic acids, or pharmacological agents into individual neurons, supporting studies of morphology, connectivity, gene function, and cellular physiology. By linking targeted interventions with observed neuronal responses, intracellular injection helps clarify mechanisms underlying neural circuits and brain function.

In this protocol a method to measure intracellular protein refolding after heat shock is described. This method can be used to study foldases like molecular chaperones and their co-factors or compounds able to influence their activity. Firefly luciferase activity is used as reporter to measure chaperone refolding activity.

Infection of Zebrafish Embryos with Intracellular Bacterial Pathogens

Transparent zebrafish embryos have proved useful model hosts to visualize and functionally study interactions between innate immune cells and intracellular bacterial pathogens, such as Salmonella typhimurium and Mycobacterium marinum. Micro-injection of bacteria and multi-color fluorescence imaging are essential techniques involved in the application of zebrafish embryo infection models.

Monitoring Changes in the Intracellular Calcium Concentration and Synaptic Efficacy in the Mollusc Aplysia

We demonstrate how changes in the intracellular free calcium concentration and synaptic efficacy can be simultaneously monitored in a ganglion preparation of Aplysia. We image intracellular calcium using a fluorescent dye, Calcium Orange, and induce and monitor synaptic transmission with sharp (intracellular) electrodes.
