Multiplex Assay

A multiplex assay is an analytical method that measures multiple biological targets simultaneously in a single sample, improving efficiency and preserving limited specimens. It works by using target-specific reagents, such as antibodies, probes, or primers, paired with distinguishable signals so each analyte can be identified and quantified without cross-interference. In neuroscience, multiplex assays can profile neurotransmitters, proteins, nucleic acids, or inflammatory markers across complex brain and nervous-system samples. These measurements help researchers compare molecular pathways, characterize disease-related changes, evaluate treatment responses, and link cellular signaling patterns to neural function.

Multiplex fluorescence immunoassays are becoming a reference for serology testing. Their application, however, is limited by the complexity and time-consuming bead-analyte coupling processes. This report presents a semi-automated, reproducible multiplex microsphere bead coupling protocol that is comparable to the standard method while considerably reducing time to results.

Analysis of tear film cytokines helps in studying various ocular diseases. Bead based multiplex assays are simple and sensitive and enable the testing of multiple targets in samples with small volumes. Here we describe a protocol for tear film cytokine profiling a using bead based multiplex assay.

Multiplex assays can provide beneficial information for basic cellular mechanisms and eliminate waste of reagents and unnecessary repetitive experiments. We describe here a multiplex caspase-3/7 activity assay, using fluorescent- and luminescent-based methods, to determine cell viability in an in vitro hypothalamic model following oxidative challenge with palmitic acid.
