Murine Stem Cell Isolation

Murine stem cell isolation is the laboratory process of recovering stem cells from mouse tissues or embryos for controlled study, making it an important foundation for neuroscience research. Tissue is mechanically and enzymatically dissociated, filtered, and separated to produce a cell suspension, after which culture conditions or cell-surface markers help enrich the desired stem cell population while preserving viability and self-renewal. Isolated cells can then be expanded, induced to differentiate into neural lineages, and analyzed for developmental, cellular, or molecular changes. This approach supports studies of neurogenesis, nervous system development, neurological disease mechanisms, and potential regenerative strategies.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Isolation of Murine Embryonic Hemogenic Endothelial Cells

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.

An optimized protocol for the isolation of stem cells from the mouse salivary gland is described. The method employs enzymatic and mechanical digestion, and permits isolation of salispheres containing cells with characteristics of stem cells.
