Nerve Injury Induction

Nerve injury induction is the controlled creation of nerve damage in an experimental model to investigate how the nervous system responds to injury and repairs itself. Researchers use defined physical or surgical interventions, such as compression, crush, or transection, to produce reproducible changes in axons, Schwann cells, neurons, and surrounding immune tissue. These models enable analysis of Wallerian degeneration, neuroinflammation, pain signaling, and axonal regeneration over time. In neuroscience, nerve injury induction supports evaluation of injury mechanisms, functional recovery, and potential therapies for peripheral neuropathy, traumatic nerve damage, and other disorders involving impaired neural connectivity.

Source: Svientek, S. R., et al. Fabrication of the Composite Regenerative Peripheral Nerve Interface (C-RPNI) in the Adult Rat. J. Vis. Exp. (2020).This video demonstrates the surgical construction of a composite regenerative peripheral nerve interface (C-RPNI) in a rat model of peripheral nerve injury. The procedure involves attaching a transected peroneal nerve to a muscle graft and overlaying it with a dermal skin graft to promote nerve regeneration. Over time, motor and sensory neurons...

Axonal Regeneration in the Sciatic Nerve of a Mouse Following a Crush Injury

Source: Li, Q., et al. Investigating Mammalian Axon Regeneration: In Vivo Electroporation of Adult Mouse Dorsal Root Ganglion. J. Vis. Exp. (2018)This video demonstrates the process of axonal regeneration in a mouse sciatic nerve following a crush injury. The isolated nerve is fixed and prepared for imaging, with fluorescent proteins expressed in neurons to visualize axon growth. Using an epifluorescence microscope, axon regeneration is traced from the injury site to assess the extent of...

The Spared Nerve Injury (SNI) Model of Induced Mechanical Allodynia in Mice

2011

The Spared Nerve Injury animal model is described here as a mouse model of peripheral neuropathic pain following partial denervation of the sciatic nerve by lesioning the tibial and common peroneal nerve branches, leaving the remaining sural nerve intact. Behavioral modification resulting from mechanical allodynia is quantified by von Frey filaments.

Inducing Fibular Nerve Injury in a Mouse to Study Neuromuscular Junction Regeneration

Source: Dalkin, W., et al. The Fibular Nerve Injury Method: A Reliable Assay to Identify and Test Factors That Repair Neuromuscular Junctions. J. Vis. Exp. (2016)This video demonstrates a surgical procedure to induce fibular nerve injury in a mouse to study neuromuscular junction (NMJ) regeneration. The procedure involves exposing the fibular nerve, applying controlled pressure to induce injury, and ensuring the epineurium remains intact to support axonal regeneration. Over time, macrophages...
