Point By Point Drilling

Point-by-point drilling is a controlled neurosurgical technique that removes cranial bone at a series of closely spaced points to create precise access to the brain. Using a fine drill, the operator advances incrementally along a marked path while limiting pressure, vibration, and heat that could damage surrounding tissue. In neuroscience, this approach supports the creation of burr holes or cranial windows for electrode implantation, neural recording, drug delivery, and imaging in animal models. Its precision improves access to targeted brain regions and can help reduce surgical trauma, making it valuable for studies of brain circuits, disease mechanisms, and experimental therapies.

Intervertebral disc degeneration is a significant contributor to back pain and a leading cause of disability worldwide. Numerous animal models of intervertebral disc degeneration exist. We demonstrate an ovine model of intervertebral disc degeneration, utilizing a drill bit, which achieves a consistent disc injury and reproducible level of disc degeneration.

Synthesis of Masarimycin, a Small Molecule Inhibitor of Gram-Positive Bacterial Growth

A detailed protocol is presented for preparing the bacteriostatic diamide masarimycin, a small molecule probe that inhibits the growth of Bacillus subtilis and Streptococcus pneumoniae by targeting cell wall degradation. Its application as a chemical probe is demonstrated in synergy/antagonism assays and morphological studies with B. subtilis and S. pneumoniae.

Accessing the Porcine Brain via High-Speed Pneumatic Drill Craniectomy

This protocol describes performing a craniectomy using a high-speed pneumatic drill on a 3-month-old Danish Landrace pig. The access is made through the frontal bone and reveals the ventral dura mater and underlying cerebral hemispheres. This procedure allows for access to a large portion of the pig brain.

This protocol describes the use of a surgical robot that performs semi-automated bone drilling of cranial windows and can help reduce surgeon-to-surgeon variability and partially mitigate thermal blood-brain barrier damage.
