Dofetilide Kidney Elimination

Dofetilide kidney elimination is the renal clearance of dofetilide, an antiarrhythmic drug, and it is clinically important because kidney function strongly influences drug exposure and safety. The kidneys remove dofetilide primarily through renal excretion, including filtration and active tubular secretion, so reduced renal function can slow clearance and increase plasma concentrations. Pharmacology studies use this relationship to guide dose selection according to creatinine clearance, assess drug interactions that may inhibit renal secretion, and reduce the risk of excessive QT prolongation and torsades de pointes. Understanding renal elimination therefore supports safer initiation, monitoring, and individualized treatment with dofetilide.

Kidney Structure

2019

The kidneys are two large bean-shaped organs located in the upper abdomen. They filter the blood several times a day to remove toxins and rebalance water and electrolytes of the circulatory system via the renal veins. The kidneys receive blood directly from the heart via the renal arteries. These arteries enter the kidney at the hilum, the concave surface of the bean, where they branch and divide into smaller vessels and capillaries. The renal cortex is the thick outer layer of the kidney. It...

In this video, we demonstrate the vibratome sectioning of a whole mouse kidney to obtain thin slices. These sliced tissue sections can be stored at low temperatures until further downstream analysis.

Enhanced Elimination of Poison

Poison can be effectively removed from the gastrointestinal (GI) tract through various decontamination procedures. Antidotes serve a crucial role in counteracting the effects of poison by inhibiting enzymes responsible for producing harmful drug metabolites. In some cases, these toxic metabolites can be neutralized by endogenous cosubstrates, which are maintained at specific concentrations to prevent interaction with cellular macromolecules and subsequent cell death. Renal excretion is the...
