Mouse Lung Epithelial Cells

Mouse lung epithelial cells are the specialized cells lining the airways and alveoli, where they maintain the respiratory barrier and support gas exchange, making them valuable models for lung biology and bioengineering. In culture, these cells establish polarized layers with distinct apical and basal surfaces, connected by cell-cell junctions and influenced by extracellular matrix signals that regulate barrier function, repair, and differentiation. Researchers use mouse lung epithelial cells to construct in vitro lung models, investigate tissue injury and regeneration, evaluate biomaterials, and study how engineered environments affect respiratory tissue behavior. Their use supports the development of more physiologically relevant platforms for disease research and therapeutic testing.

Source: Li, T., et al. Studying Effects of Cigarette Smoke on Pseudomonas Infection in Lung Epithelial Cells. J. Vis. Exp. (2020).This video demonstrates how cigarette smoke extract weakens antimicrobial defense in lung epithelial cells by reducing AMP production, leading to increased bacterial invasion. The procedure measures intracellular bacterial load to assess the impact of smoke exposure on host-pathogen interactions.

2025

This protocol presents a flow cytometry-based method to isolate viable alveolar type II epithelial cells (AECII) from neonatal, juvenile, or adult murine lungs (infected or non-infected). This approach enables downstream molecular and/or functional studies of AECII to identify their roles during lung inflammation and infection.
