Peyer Patches

Peyer’s patches are organized lymphoid tissues in the small intestine that monitor intestinal contents and coordinate mucosal immune defense. Specialized microfold, or M, cells transport antigens from the gut lumen to underlying immune cells, where antigen-presenting cells activate T cells and B cells; stimulated B cells can support production of immunoglobulin A. This process helps the body distinguish harmful microbes from harmless dietary and resident microbial components while limiting unnecessary inflammation. Studying Peyer’s patches provides insight into intestinal homeostasis, infection, oral tolerance, inflammatory disease, and the design of vaccines or drug-delivery systems that target mucosal immunity.

Here we outline parallel protocols for preparing PP single cell preparations for flow cytometric analysis and PP cryosections for immunostaining.

Here, we describe a precise method for collecting thoracic duct lymphocytes and observing the migration of gut-tropic lymphocytes in rat Peyer’s patches for 3 hours using time-lapse photography. This technique can clarify how the dynamics of lymphocytes are affected under inflammatory conditions.
