In Vitro Protocol

An in vitro protocol is a controlled laboratory procedure performed outside a living organism to investigate biological, chemical, or material-based processes under defined conditions. It typically specifies the samples, reagents, equipment, environmental parameters, and sequential steps needed to produce reproducible results, while controlling factors such as temperature, pH, nutrient availability, and exposure time. In engineering, in vitro protocols support the evaluation of biomaterials, medical devices, engineered tissues, and cell-material interactions before further testing. By standardizing experiments and enabling direct measurement of responses, these protocols help researchers compare designs, identify performance limits, and refine safer, more effective technologies.

Testing protein-protein interaction is indispensable for dissection of protein functionality. Here, we introduce an in vitro protein-protein binding assay to probe a membrane-immobilized protein with a soluble protein. This assay provides a reliable method to test interaction between an insoluble protein and a protein in solution.
