Cholesterol Doped Bicelles

Cholesterol-doped bicelles are nanoscale, disk-shaped lipid assemblies that combine a phospholipid bilayer region with detergent or short-chain-lipid edges, while incorporated cholesterol tunes their membrane-like properties. They form through the self-assembly of amphiphilic molecules in aqueous solution; cholesterol inserts between lipid molecules, altering chain packing, fluidity, permeability, and structural stability as composition and temperature change. In engineering research, these assemblies provide controllable model membranes for studying interfacial organization, membrane-protein behavior, and transport, and they can support the design of biosensors, drug-delivery systems, and other lipid-based nanomaterials.

In this article, a protocol for infection of macrophages with Cryptococcus neoformans is described. Also, a method for sterol depletion from the macrophages is explained. These protocols provide a guide to study fungal infections in vitro and examine the role of sterols in such infections.

A detailed procedure for surface doping of Silicon interfaces is provided. The ultra-shallow surface doping is demonstrated by using phosphorus containing monolayers and rapid annealing process. The method can be used for doping of macroscopic area surfaces as well as nanostructures.
