Blood Stimulation

Blood stimulation is an ex vivo laboratory method that activates immune cells in whole blood or blood-derived samples to examine host responses to infection, inflammation, or immunomodulatory signals. In a typical assay, microbial antigens, pathogen-associated molecular patterns, or other stimuli engage receptors on leukocytes, triggering intracellular signaling and the release of cytokines, chemokines, and other immune mediators. Researchers measure these responses using immunoassays, flow cytometry, or gene-expression analyses to characterize immune function and compare responses across conditions. Blood stimulation supports infection research, immune profiling, vaccine evaluation, and investigation of factors that regulate innate and adaptive immunity.

Accurate and Simple Measurement of the Pro-inflammatory Cytokine IL-1β using a Whole Blood Stimulation Assay

We describe a simple immunoassay to measure the production of pro-inflammatory cytokines, such as IL-1 beta production, in patients presenting with autoinflammatory phenotypes. By activating cells in whole blood cultures with pathogen-associated molecular patterns, specifically with lipopolysaccharide, cytokine secretion can be conveniently evaluated in whole blood supernatants.

Induction of Alloantigen-specific Anergy in Human Peripheral Blood Mononuclear Cells by Alloantigen Stimulation with Co-stimulatory Signal Blockade

This paper describes a simple technique to induce alloantigen-specific anergy in human peripheral blood mononuclear cells. The technique can be applied clinically to generate non-alloreactive donor cells. Infusion of these cells could improve immune reconstitution and reduce toxicity after allogeneic hematopoietic stem cell transplantation.

Improved Method for the Preparation of a Human Cell-based, Contact Model of the Blood-Brain Barrier

Establishment of human models of the blood-brain barrier (BBB) can benefit research into brain conditions associated with BBB failure. We describe here an improved technique for preparation of a contact BBB model, which permits coculturing of human astrocytes and brain endothelial cells on the opposite sides of a porous membrane.

Transcranial magnetic stimulation (TMS) has proven to be a useful tool in investigating the role of the articulatory motor cortex in speech perception. This article describes how to record motor evoked potentials (MEPs) from the lip muscles and how to disrupt the motor lip representation using repetitive TMS.
