Rbl-2h3 Cells

RBL-2H3 cells are a rat basophilic leukemia cell line widely used as an in vitro model of mast cell and basophil activation in immunology. These cells express the high-affinity IgE receptor FcεRI; when antigen cross-links receptor-bound IgE, intracellular signaling triggers granule exocytosis and the release of mediators such as histamine and β-hexosaminidase. Researchers use RBL-2H3 cells to measure degranulation, analyze Fc receptor signaling, and evaluate allergens, antibodies, and other immune-modulating compounds. Their consistent responses provide a practical system for studying immediate hypersensitivity and mast cell contributions to inflammatory and host-defense responses.

Here we demonstrate techniques1 for assessing effects of drugs and toxicants on degranulation, methodology recently utilized to exhibit the powerful inhibitory effect of antibacterial agent triclosan2.

An In Vitro Technique to Visualize Exocytosis of Secretory Granules in Mast Cells

This video demonstrates a technique to visualize exocytosis in mast cells. The cells are incubated with FITC-dextran — a fluorescent reporter — which is taken up by cells inside secretory granules (SGs), where the acidic pH quenches its fluorescence. Upon stimulation of granule exocytosis, the medium's higher pH induces reporter fluorescence inside the fused SGs, which is visualized using a fluorescence microscope.

In this video, we describe an in vitro method to evaluate the allergic potential of an antigen to induce basophil degranulation. β-hexosaminidase is used as an index of chemical mediator release, and the enzyme activity of β-hexosaminidase is measured using a fluorogenic substrate.
