Sars Vaccine

SARS vaccines are preventive immunizations designed to prepare the immune system to recognize and control SARS-CoV, the coronavirus that causes severe acute respiratory syndrome, before infection occurs. They present viral antigens, such as the spike protein or genetic instructions for producing it, so antigen-presenting cells activate B and T lymphocytes; B cells can generate neutralizing antibodies that block viral entry, while T cells support infected-cell clearance and immune memory. Research on SARS vaccine platforms helps evaluate protection, durability, safety, and potential immune enhancement, while informing coronavirus vaccine design and preparedness for related emerging pathogens.

Protocol for Recombinant RBD-based SARS Vaccines: Protein Preparation, Animal Vaccination and Neutralization Detection

This protocol describes a general procedure for studying recombinant receptor-binding domain (RBD)-based subunit vaccines against SARS. It includes methods for transfection and expression of RBD protein in 293T cells, immunization of mice with RBD and detection of neutralization activity of mouse sera using an established SARS pseudovirus neutralization assay.

Prophylactic and therapeutic vaccination often fails to stimulate strong immune responses due to week drainage of the vaccine to lymph nodes and consequently poor involvement of immune cells. By direct injection of vaccine to lymph nodes, so-called intralymphatic injection, vaccine efficacy can be strongly improved and vaccine doses can be reduced.
