Serotype Concordance

Serotype concordance is the degree to which two tests, laboratories, specimens, or classification systems assign the same serotype to an infectious agent, providing a measure of reliability in immunology and infection research. It is assessed by comparing serotype calls, often generated through antibody-based reactions or molecular typing of antigen-associated markers, and summarizing matching results as percentage agreement or statistics such as Cohen’s kappa. Researchers use it to validate diagnostic assays, monitor circulating strains, and support consistent epidemiological conclusions. High concordance strengthens confidence in surveillance, vaccine studies, and analyses linking serotype-specific immune responses to infection risk, while discordance can reveal assay limitations or antigenic variation.

Latex agglutination testing is a simple, rapid and inexpensive method for serotyping Streptococcus pneumoniae, and has also been widely applied in diagnostic microbiology. This manuscript describes the in-house production of latex agglutination reagents, quality control procedures and the application of this technique to pneumococcal serotyping.

Here, we present a protocol to generate a proof-of-principle divalent adenovirus type 5 (Ad5) vector Ad5/H5-HVR1-KWAS-HVR5-His6 by utilizing the Antigen Capsid-Incorporation strategy. This vector was demonstrated to exhibit qualitative fitness, the capability to escape Ad5-positive sera in vitro, and the antigenicity as well as immunogenicity to the incorporated antigens.

Systemic Delivery of MicroRNA Using Recombinant Adeno-associated Virus Serotype 9 to Treat Neuromuscular Diseases in Rodents

Here we describe the delivery of microRNA using a recombinant adeno-associated virus serotype 9 in a mouse model of a neuromuscular disease. A single peripheral administration in mice resulted in sustained miRNA overexpression in muscle and motor neurons, providing an opportunity to study miRNA function and therapeutic potential in vivo.
