Viral Nucleocapsid

A viral nucleocapsid is the nucleic acid genome together with the protein shell or nucleoprotein complex that packages it inside a virus, helping organize genetic material and shape infection. Its structure depends on specific interactions between viral RNA or DNA and capsid or nucleocapsid proteins; these interactions can protect the genome, support particle assembly, and regulate genome release or replication. In immunology and infection research, nucleocapsid components are important targets for antibody and T-cell responses, diagnostic assays, and antiviral studies. Comparing nucleocapsid structure and antigenicity across viruses can clarify transmission, immune recognition, and opportunities for vaccine or therapeutic development.

We describe the process of isolating high purity herpesvirus nucleocapsid DNA from infected cells. The final DNA captured from solution is of high concentration and purity, making it ideally suited for high-throughput sequencing, high fidelity PCR reactions, and transfections to produce new viral recombinants.

Viral Structure

Viruses are extraordinarily diverse in shape and size, but they all have several structural features in common. All viruses have a core that contains a DNA- or RNA-based genome. The core is surrounded by a protective coat of proteins called the capsid. The capsid is composed of subunits called capsomeres. The capsid and genome-containing core are together known as the nucleocapsid. Structural Classes of Viruses Many criteria are used to classify viruses, including capsid design. Most viruses...
