Chronic Constriction Injury

Chronic constriction injury (CCI) is an experimental model of peripheral neuropathic pain in which a peripheral nerve is loosely ligated to produce persistent sensory abnormalities. The constriction partially compresses the nerve, triggering local inflammation, altered axonal signaling, and abnormal spontaneous activity that can generate mechanical allodynia and thermal hyperalgesia. In medicine and pain research, CCI is used primarily in laboratory animals to investigate the mechanisms of nerve injury and evaluate analgesic or neuroprotective treatments. Findings from this model help clarify how peripheral nerve damage contributes to chronic pain, while also supporting comparisons with other neuropathic pain models.

Chronic Constriction of the Sciatic Nerve and Pain Hypersensitivity Testing in Rats

Due to the simplicity of surgery and the robust behavioural outcome, chronic constriction of the sciatic nerve is one of the pre-eminent animal models of neuropathic pain. Within 24 hrs following surgery, pain hypersensitivity is established and can be quantitatively measured using a von Frey aesthesiometer (mechanical test) and plantar analgesia meter (thermal test).

This manuscript describes a rodent model to study trigeminal neuropathic pain. The methods described include surgical procedures to perform a chronic constriction injury of the rat’s infraorbital nerve and the post-surgical behavioral tests to measure the changes in spontaneous and evoked behavior that are indicative of persistent pain and allodynia.

Ascending Aortic Constriction in Rats for Creation of Pressure Overload Cardiac Hypertrophy Model

We describe a stepwise procedure for creating pressure overload and left ventricular hypertrophy in Wistar rats by constriction of the ascending aorta using a small metallic clip. This model is extensively used for studying remodeling changes during cardiac hypertrophy and for identifying and evaluating strategies for regression of such changes.
