Embryonic Day 18

Embryonic Day 18 (E18) is a standardized developmental time point, most commonly referring to 18 days after fertilization in mouse research, when late-gestation embryos undergo substantial organ maturation. Researchers identify this stage by timed mating and calculate embryonic age from fertilization or the appearance of a copulation plug, allowing developmental changes to be compared across experiments. At E18, coordinated cell differentiation, tissue remodeling, and functional maturation provide a defined context for examining anatomy and physiology. In medicine, E18 models support studies of fetal development, congenital disorders, neurobiology, prenatal toxicology, and the effects of genetic or pharmacological interventions.

Here, we describe a method for isolation, culture and manipulation of mouse embryonic pancreas. This represents an excellent ex vivo system for studying various aspects of pancreatic development, including morphogenesis, differentiation and growth. Pancreatic bud explants can be cultured for several days and used in a range of different applications, including whole-mount immunofluorescence and live imaging.

A facile, one-pot synthesis of N-succinimidyl-4-[18F]fluorobenzoate ([18F]SFB) was developed based on a non-aqueous, three-step radiochemical process. Using microwave heating, the entire procedure can be completed in less than 30 min, or 60 min with further purification by preparative HPLC. The decay-corrected radiochemical yields (RCYs) were 35-5% (n > 30).

Stable isotope labeling workflows employing 18O-enriched water (LeO-workflows) are versatile tools for quantitative and qualitative proteomics studies. In protease-assisted (PALeO) workflows, 18O-atoms are introduced by proteolytic cleavage and carboxyl oxygen exchange reactions mediated by proteases. In the acid-catalyzed (ALeO) workflow, 18O-atoms are introduced by carboxyl oxygen exchange at low pH.
