Splenic Lymphocyte Migration

Splenic lymphocyte migration is the regulated movement of immune cells within and through the spleen, a process that supports immune surveillance and coordinated antigen responses. Chemokine gradients, adhesion molecules, and signals from stromal cells guide lymphocytes between the blood, white pulp, marginal zone, and red pulp, where they encounter antigens or undergo recirculation. T and B lymphocytes use distinct trafficking cues to reach specialized microenvironments and interact with antigen-presenting cells. Understanding this movement helps explain how the spleen filters blood, organizes adaptive immunity, and contributes to inflammatory, infectious, and autoimmune disease.

Assessment of Lymphocyte Migration in an Ex Vivo Transmigration System

In this protocol, lymphocytes are placed in the top chamber of a transmigration system, separated from the bottom chamber by a porous membrane. Chemokine is added to the bottom chamber, which induces active migration along a chemokine gradient. After 48 h, lymphocytes are counted in both chambers to quantitate transmigration.

Here, we present a lymphocyte purification protocol to determine the physiological functions of the desired molecules in lymphocyte activation in vitro and in vivo. The described experimental procedures are suitable for comparing functional capacities between control and genetically modified lymphocytes.
