T Cell Subsets

T cell subsets are functionally distinct populations of T lymphocytes that coordinate immune defense, regulate inflammation, and establish immunological memory. They arise when naïve T cells recognize antigen through the T cell receptor and receive specific cytokine and costimulatory signals, directing differentiation into helper, cytotoxic, regulatory, or memory populations with specialized gene-expression programs and effector functions. In medicine, characterizing subsets such as CD4+ helper, CD8+ cytotoxic, regulatory, and memory T cells helps explain infection control, autoimmunity, immunodeficiency, and tumor immunity. Subset profiling also supports vaccine development, immune monitoring, disease diagnosis, and the design of targeted cellular immunotherapies.

Isolation of Precursor B-cell Subsets from Umbilical Cord Blood

Here we describe a protocol for isolating subsets of precursor B-cells from umbilical cord blood. A sufficient quantity and quality of nucleic acids may be extracted from the cells and used in subsequent assays utilizing DNA or RNA.

Naïve CD4+ T cells polarize to various subsets depending on the environment at the time of activation. The differentiation of naïve CD4+ T cells to various effector subsets can be achieved in vitro through the addition of T cell receptor stimuli and specific cytokine signals.

Distinct dendritic cell subsets exist as rare populations in lymphoid organs, and therefore are challenging to isolate in sufficient numbers and purity for immunological experiments. Here we describe a high efficiency, high yield method for isolation of all of the currently known major subsets of mouse splenic dendritic cells.

This video demonstrates a flow cytometry-based technique using two fluorochromes to identify CD4+ and CD8+ T cells, γδ T cells, B cells, natural killer (NK) cells, and monocytes in human peripheral blood mononuclear cells (PBMCs). Upon incubating the cells with fluorochrome-conjugated antibodies against the surface markers, the cell subsets are identified using an optimal flow cytometric gating strategy.
