Neuron Macrophage Coculture

Neuron macrophage coculture is an in vitro model that grows neurons and macrophages together to study communication between the nervous and immune systems. In this system, macrophages respond to neuronal signals and environmental cues by changing their activation state, while neurons can be affected by macrophage-derived cytokines, growth factors, and inflammatory mediators. Researchers use direct-contact or compartmentalized cultures to distinguish effects requiring cell contact from those mediated by soluble factors. These models help investigate neuroinflammation, neuronal injury and repair, neurodegenerative disease mechanisms, and immune responses in the nervous system, providing a controlled platform for testing how immune modulation influences neuronal survival and function.

Developing a Neuron and Macrophage Coculture In Vitro

2025

This video shows the co-culturing of murine dorsal root ganglion neurons and macrophages. Neurons are seeded on a coated plate, while macrophages are collected from an euthanized mouse, treated, and added to the culture insert. Treatment with db-cAMP activates neurons and prompts macrophages to adopt a pro-regenerative phenotype to support neuronal outgrowth.

Coculture Assays to Study Macrophage and Microglia Stimulation of Glioblastoma Invasion

Understanding the malignant behavior of cancer requires creating accurate models of how tumor cells interact with components of the tumor microenvironment, such as macrophages. Here we describe two methods to study glioblastoma cell interaction with tumor associated macrophages and microglia where the effect on glioblastoma invasion is assessed.

This video demonstrates the procedure to isolate retinal ganglion neurons (RGNs) from a rat's retina and coculture them with olfactory ensheathing glia (OEG) cells. This method allows for studying the neuro-regenerative potential of OEGs after neural injury.

Using a Macrophage Conditioned Medium to Promote Neuron Extensions

This video demonstrates an assay for promoting the growth of neuron projections known as neurite outgrowth using the macrophage-conditioned medium. Co-culture neurons and macrophages with cAMP and harvest the macrophage-derived conditioned medium. Growing the neurons with the conditioned medium promotes neurite outgrowths. These outgrowths are fluorescently labeled and visualized using a fluorescence microscope.
